How long do you leave a mushroom for a spore print? ›

Maintaining the ideal humidity is perhaps the trickiest part of making spore prints. Dry mushrooms won't give a good spore print, but wet ones can make a sloppy mess. If you put a small mushroom on a piece of paper and cover it with a large bowl you may end up with a shrivelled cap and no spore print.

Colonization occurs after inoculation. This is when the mycelium will start to take over the grain that was inoculated with spores. This period usually lasts between 3 to 6 weeks depending on strain and environmental conditions. Ideal conditions for this phase are in a dark place and temperature between 72 F – 80 F.

Dry is very important, as moisture “activates” mushroom spores, meaning they will have a significantly shorter lifespan. Keeping conditions airtight is important as well. Not only will this allow your prints to remain viable for longer, but it will also keep them sterile too!

What color is a shiitake spore print? ›

To reliably distinguish a Galerina from a shiitake, make a spore print. Cut off the stem and place the cap, gills down, on white paper. Cover it with a bowl to keep it moist. Galerina always gives a brown spore print after some hours to overnight; a shiitake spore print is always white.

Scrape spores from the print into the sterile syringe carefully. Add the cooled, boiled water to the syringe. Use just enough to mix with the spores. Shake the syringe gently.

The spore prints are permanent. The illustra- tion shows a spore print from Gymnopilus junonius made in 1988 which still retains its colour. The spores can be sampled but this entails a degree of destruction.

Colonization from a spore syringe will take anywhere from 2-6 weeks depending on how much grain is in your container, how much spore solution you inoculate with, and how optimized you are able to make the environmental conditions. Colonization from a Liquid Culture syringe will take anywhere from 2-4 weeks.

Remove the stem of the specimen and place the cap of the mushroom on the spore print card with the gills (or pores) facing downward. Cover it or leave it undisturbed overnight and compare the colour of the resulting print with descriptions in your field guide.

The majority will germinate within 5 minutes after exposure to a high level of an appropriate germinant [172]. The remainder will typically follow suit after another 15 minutes, yet there are superdormant spores that are capable of significantly delaying germination for hours to days [173].

A mushroom cannot be identified from its spore print alone; the spore print is only one characteristic used in making a taxonomic determination. Spore prints are usually white to cream, black, or shades of red, purple, or brown. The poisonous false parasol (Chlorophyllum molybdites) has a green spore deposit.
